The presentation looks at the role of flocculation in the recovery and purification of monoclonal antibodies through direct experiments and case studies. Presented examples show how in situ technologies such as Focused Beam Reflectance Measurement (FBRM) can help optimize flocculation for easier, more effective downstream processing.
Flocculation of cells and cell debris has emerged as a useful pre-treatment for clarification of cell culture separation. However, as cell densities increase in the hunt for higher-producing titers, the result is often more impurities and broader size distributions which negatively impact cell viability and downstream processing.
In situ PAT is giving researchers and manufacturers a continuous view on the flocculation process to help them improve efficiency and create higher-recovery yields, even for high-density solid-liquid separations.
